Research Proposal: The current proposal will (1) determine the effect of mutation of each residue of the N-terminal cytoplasmic tail on the Golgi localization of GlcNAc-1-phosphotransferase (2) Search for a panel of replacement lysosomal enzymes that might provide benefit for ML patients (3) Investigate the molecular basis for increased c-Met receptor phosphorylation in GNPTAB-null cells and (4) Assess that pathogenic contributions of individual proteases dysregulated in ML2 and explore the efficacy of individual pharmacological inhibitors to reduce their activity. This information may point to new ways to treat ML disease.
